The present invention relates to a method for providing .alpha.-glucosidase inhibition to a subject by administering a pharmaceutical composition comprising a .alpha.-glucosidase inhibitory agent selected from pipataline (formula 1a), sesamin (formula 1b), pellitorine (Formula 1c), guineensine (Formula 1d) and brachystamide-B (formula 1e) having therapeutic application for diabetes mellitus, cancer, viral diseases such as hepatitis B and C, HIV, AIDS etc; also the invention provides a process for the isolation of said .alpha.-glucosidase inhibitory agent from the plant source Piper longum in significant yields.

Isomaltooligosaccharides (IMOs) produced by Leuconostoc mesenteroides ATCC 13146 fermentation with a sucrose:maltose ratio of 2:1 have been discovered to be effective prebiotics in mixed cultures of microbial populations, including cultures from chicken ceca. Surprisingly in mixed microbial cultures this IMO composition proved as effective as FOS. Thus, these IMOs can be used as effective prebiotics for both birds and mammals. Moreover, the IMOs were discovered to be effective non-competitive inhibitors of .alpha.-glucosidase. These IMOs also will be useful, as an .alpha.-glucosidase inhibitor, in a therapeutic application for several diseases, including obesity, diabetes mellitus, pre-diabetes, gastritis, gastric ulcer, duodenal ulcer, caries, cancer, viral disease such as hepatitis B and C, HIV, and AIDS. A diet with 5-20% IMOs was also shown to reduce the abdominal fat tissue in mammals.

A method of regulating oncogene-mediated cell transformation in a mammalian host. Oncogenes having glycosylated expression products are regulated by administering an effective amount of a processing glucosidase inhibitor: a glucosidase I inhibitor, e.g., castanospermine, N-methyl-1-deoxynojirimycin, 1-deoxynojirimycin, 5-amino-5-deoxy-D-glucopyranose; or a glucosidase II inhibitor, e.g., bromoconduritol. The glucosidase I inhibitors are preferred, particularly castanospermine (CA) and N-methyl-1-deoxynojirimycin (MdN). Oncogenes having glycosylated expression products that are ultimately expressed on the plasma membrane or secreted from transformed cells are particularly susceptible to regulation by these anti-cancer drugs. Also provided is a method of regulating the immune system of a mammalian host. Administration of an effective amount of a processing glucosidase inhibitor suppresses proliferation and differentiation of monocytic and myeloblastic cells.
