Immunochemical assays for human amylase isoenzymes are disclosed. These assays utilize monoclonal antibodies which are produced from hybridoma cell lines and are specific for human salivary amylase and ovarian tumor amylase but not human pancreatic amylase. The assays involve reacting this monoclonal antibody with the unknown sample for the purpose of studying quantitatively and qualitatively the amylase isoenzymes present. The results from such assays are useful in the diagnosis of disease or ruling out disease. Antibody producing hybridoma cell lines and their production are described.

The invention is directed to a pharmaceutical preparation with a modulatory effect on malignant tumors, which contains a combination of protease proenzymes and amylases in a ratio between 1:100 and 100:1 in enzymatic activity units, and a protease inhibitor. The invention is also directed to use of such a preparation to modulate the effects of malignant tumors in humans and animals.

A gene expression system is used to produce heterologous biologically active proteins, in particular bioactive granulocyte macrophage colony stimulating factor ("GM-CSF"), secreted from a host selected from the Streptomyces genera. Novel signal peptides of these constructs include hybrids of the signal peptides of Streptomyces griseus protease B and Escherichia coli omp A, hybrids of Streptomyces griseus protease B and Streptomyces limosus .alpha.-amylase signal peptide.
